Flow-based solid phase peptide synthesis (SPPS) has enabled the chemical synthesis of long peptides and single domain proteins in a few hours of synthesis time.[1] A rapid, automated flow chemistry platform allows for the collection of time-resolved synthesis data and gives access to information that is inaccessible with traditional synthesis methods. This analytical data can be used to further optimize and predict chemical synthesis outcome, including sequence-dependent events such as aggregation.[2] This technology holds the promise to advance current peptide synthesis methods and to produce chemically modified peptides and proteins on demand. However, challenges such as low atom economy and the occurrence of sequence-specific events -such as aggregation- remain unsolved. In this presentation, recent advances in flow-based peptide synthesis will be highlighted and strategies to render the technology more sustainable will be presented.
